Major life changes following psychedelic use: A retrospective survey among people using psychedelics naturalistically

This survey (n=581) evaluates the Psychedelic-related Major Life Changes Questionnaire (P-MLCQ) in people reporting naturalistic psychedelic use. It finds that 82.96% of participants reported major life changes in at least one domain, including goals (53.7%), values (53.53%), and spirituality (49.05%), with changes rated highly positively (M = 4.64/5). Frequency of use correlated with more changes (r = 0.34), while education level was negatively associated with the number of changes (β = -0.137).

Mixed-methods analysis on psychedelic-augmented meditation experiences from a randomized controlled mindfulness retreat

This secondary analysis of a randomised controlled trial of DMT/harmine in experienced meditators found that meditation under DMT-harmine differed from placebo in its thematic and experiential profile while still sharing some semantic features. Using NLP-based analysis of phenomenological interviews, the study also suggested overlap between meditative and psychedelic states and highlighted the strong influence of context and spiritual framing on subjective experience.

Pilot study of psilocybin in patients with post-treatment lyme disease

In an open‑label, single‑arm pilot study of 20 patients with post‑treatment Lyme disease, two psilocybin sessions with psychological support produced significant, sustained reductions in symptom burden (40% decrease in GSQ‑30 at six months) and improvements in both physical and mental quality of life. The intervention was feasible and well-tolerated with no serious related adverse events, supporting further controlled trials of psilocybin‑assisted therapy for PTLD.

Inhaled N, N-dimethyltryptamine diminishes connectivity between the ventral tegmental area and the nucleus accumbens: relevance to pathologies of mesolimbic and mesocortical pathways

In a within-subject pharmacoimaging study of 11 healthy psychedelic-experienced volunteers, inhaled DMT acutely reduced connectivity between the left ventral tegmental area and the right nucleus accumbens while increasing connectivity between the nucleus accumbens and anterior cingulate cortex and between the medial prefrontal cortex and anterior cingulate. These connectivity shifts correlated with changes in volition and perception and suggest a potential therapeutic mechanism for disorders of reward processing.

Acute experiences and persisting psychological effects associated with an encapsulated DMT-harmala alkaloid combination: results of a phase 1 study

This open-label study (n=9) found that an encapsulated DMT-harmala alkaloid product (pharmahuasca) produced dose-dependent mystical experiences that exceeded those reported in most previous ayahuasca studies and were associated with beneficial persisting psychological effects in healthy volunteers.

Mapping the phenomenology of intranasal 5-MeO-DMT in psychedelic-naïve healthy adults

This Phase I double-blind placebo-controlled trial (n=32) using microphenomenology interviews found that intranasal 5-MeO-DMT produced dose-dependent subjective effects with rapid onset peaking at 8-15 minutes and return to baseline by 45-60 minutes, characterised by minimal visual effects but strong emotional and bodily experiences, including emotional breakthroughs and personal insights.
