Modeled Long-Term Effects of Psilocybin on Dynamic Activity and Effective Connectivity of Fronto-Striatal-Thalamic Circuits

This secondary analysis and computational modelling study of resting-state fMRI data from a within-subject longitudinal psilocybin trial in psychedelic-naïve healthy volunteers found that fronto-striatal-thalamic circuits became more dynamically active 4 weeks after a full dose. The changes were linked to reduced top-down and increased bottom-up information flow, with receptor maps suggesting roles for 5-HT2A and D2 receptors.

Investigating Emotional Reactivity in Experienced Users of Psychedelics: a cross-sectional fMRI study

In a preregistered cross-sectional fMRI study comparing experienced naturalistic psychedelic users (≥10 lifetime uses; N = 33) with matched non‑users (N = 34), users were faster and more accurate at recognising angry faces, suggesting reduced interference from threat-related stimuli. Neuroimaging showed diminished limbic and salience‑network responses to anger, enhanced parietal and sensorimotor responses to happiness, increased precuneus activation to fear, and reduced emotional‑category differentiation in the frontal medial cortex and parahippocampal gyrus, indicating sustained neurofunctional alterations in emotional processing associated with naturalistic psychedelic use.

The Role of the Dorsolateral Prefrontal Cortex in Ego Dissolution and Emotional Arousal During the Psychedelic State

Using fMRI and MEG in healthy volunteers during acute LSD, the study shows that DLPFC functional connectivity—notably bilateral DLPFC–thalamus–FFA coupling—correlates with subjective ego dissolution, while right DLPFC–IPS–salience network coupling correlates with emotional arousal. MEG Granger causality further revealed increased theta‑band information flow between thalamus and DLPFC, consistent with disrupted thalamic gating during ego dissolution.

P300-mediated modulations in self-other processing under psychedelic psilocybin are related to connectedness and changed meaning: A window into the self-other overlap

In a placebo‑controlled, double‑blind crossover EEG study (n = 17), psilocybin abolished the P300‑timeframe distinction between self‑ and other‑related auditory feedback—driven by current source density changes in the supragenual anterior cingulate and right insula—and the magnitude of this effect correlated with subjective feelings of unity and changed meaning. These findings indicate 5‑HT‑mediated modulation of late self‑referential encoding underlies altered self–other overlap and may inform therapeutic models emphasising connectedness.

Trait Openness and serotonin 2A receptors in healthy volunteers: A positron emission tomography study

In 159 healthy volunteers, PET imaging with [18F]altanserin and [11C]Cimbi‑36 showed no significant association between neocortical 5‑HT2A receptor availability and trait Openness (NEO‑PI‑R), and no sex interactions. Thus baseline 5‑HT2A binding does not appear to account for individual differences in Openness despite evidence that 5‑HT2A agonists such as psilocybin can increase the trait.

LSD-induced entropic brain activity predicts subsequent personality change

In a placebo‐controlled resting‑state fMRI study, intravenous LSD robustly increased sample entropy across sensory and higher‑order brain networks. These LSD‑induced entropy increases—most predictive during music listening and when ego‑dissolution was reported—forecasted enduring increases in the personality trait openness at a two‑week follow‑up.
