Absolute Oral Bioavailability and Bioequivalence of LSD Base and Tartrate in a Double-Blind, Placebo-Controlled, Crossover Study

In a randomized, double‑blind, placebo‑controlled five‑period crossover study in 20 healthy volunteers, different oral LSD formulations (base and tartrate; solutions and tablet) were bioequivalent with an absolute oral bioavailability of about 80%. Intravenous LSD produced larger subjective effects and more adverse effects (anxiety, nausea, negative experiences) than oral administration.

Acute Effects and Pharmacokinetics of LSD after Paroxetine or Placebo Pre-Administration in a Randomized, Double-Blind, Cross-Over Phase I Trial

In a randomized, double‑blind, cross‑over trial in 23 healthy volunteers, daily paroxetine (a CYP2D6 inhibitor) did not change LSD's pleasant subjective effects but significantly reduced adverse effects (bad drug effect, anxiety, nausea) and increased LSD Cmax and AUC by ~1.4–1.5-fold. The findings indicate CYP2D6 contributes to LSD metabolism and suggest co‑administration with SSRIs that inhibit CYP2D6 is well tolerated and likely does not require LSD dose adjustment, although recommendations for SSRIs that do not inhibit CYP2D6 remain uncertain.

A Phase 1 Assessment of the Safety, Tolerability, Pharmacokinetics and Pharmacodynamics of (2R,6R)-Hydroxynorketamine in Healthy Volunteers

In a Phase 1 single- and multiple-ascending IV dosing study in healthy volunteers, (2R,6R)-hydroxynorketamine was well tolerated with minimal adverse events, showed no anesthetic or dissociative effects and demonstrated dose-proportional pharmacokinetics with confirmed CNS exposure in CSF. Quantitative EEG showed increases in gamma power in some participants, supporting progression of RR‑HNK into Phase 2 development.

Assessment of the acute effects of 2C-B vs psilocybin on subjective experience, mood and cognition

In a within‑subjects, double‑blind, placebo‑controlled study (n=22) of psychedelic‑experienced volunteers, 20 mg 2C‑B produced acute psychedelic alterations of moderate experiential depth but elicited less dysphoria, subjective impairment, auditory changes and ego‑dissolution than 15 mg psilocybin. Both drugs produced comparable psychomotor slowing, spatial memory impairments and transient pressor effects, while 2C‑B’s subjective effects were shorter in duration (largely resolving within ~6 hours).

Pharmacokinetics and pharmacodynamics of oral psilocybin administration in healthy participants

This study analysed data from three clinical trials (n=79) to characterize the pharmacokinetic-pharmacodynamic relationship of orally administered psilocybin (15-30 mg). Maximal psilocin concentrations were 11 ng/ml, 17 ng/ml, and 21 ng/ml after the administration of 15, 25, and 30 mg psilocybin, respectively, and maximal levels were reached after an average of 2 hours. The duration and intensity of subjective effects were dose-dependent.

Acute effects of psilocybin after escitalopram or placebo pretreatment in a randomized, double-blind, placebo-controlled, cross-over study in healthy subjects

In a randomised, double-blind, placebo-controlled crossover study in healthy volunteers, 14 days of escitalopram pretreatment attenuated psilocybin-induced bad drug effects, anxiety and adverse cardiovascular effects while not reducing positive mood effects. Escitalopram also did not alter psilocin pharmacokinetics, QTc, circulating BDNF or HTR2A/SLC6A4 expression, and the authors note further work is needed with longer pretreatment and clinical populations.
