Sustained pharmacodynamic effects of S‐ketamine on cortical excitability and resting‐state brain activity: A randomized, placebo‐controlled trial

This randomised, double-blind, double-dummy, placebo-controlled crossover trial (n=16) in healthy participants examined acute and delayed brain effects of S-ketamine given intravenously or by mouth. It found sustained changes in cortical excitability and resting-state brain activity lasting up to 7 days, especially after intravenous and high-dose oral dosing.

Safety pharmacology of acute mescaline administration in healthy participants

This pooled analysis of two double‑blind, randomized, placebo‑controlled studies (48 participants, 96 administrations) found that single oral doses of mescaline 100–800 mg produced dose‑dependent increases in positive subjective effects with only moderate autonomic changes and no clinically significant liver, kidney or haematological abnormalities. Although nausea was dose‑limiting and small proportions experienced transient diastolic hypertension, tachycardia, fever or “flashbacks”, single doses up to 800 mg were considered safe with respect to acute psychological and physical harm in healthy participants in a controlled clinical setting.

Pharmacokinetics, pharmacodynamics and urinary recovery of oral lysergic acid diethylamide (LSD) administration in healthy participants

In 28 healthy volunteers given 85 or 170 μg oral LSD, pharmacokinetic–pharmacodynamic modelling showed dose‑proportional pharmacokinetics with mean Cmax 1.8 and 3.4 ng/mL at ~1.7 h, elimination half‑lives of ~3.7–4.0 h, and dose‑dependent subjective effects lasting ~9–11 h. The drug was extensively metabolised with only ~1% recovered unchanged and ~16% as 2‑oxo‑3‑hydroxy‑LSD in urine over 24 h, validating prior findings and providing the first detailed urinary recovery data.

Ketamine for suicidality: an umbrella review

This review (2022) investigated the role of ketamine in suicidal ideation (SI) and behaviours. Of 27 studies that addressed ketamine for SI, only four reported mixed or negative results. Out of nine reviews, esketamine was significantly beneficial in five. Despite the majority of reviews being of critically low quality, the short-term efficacy of ketamine in suicidality was noted in most.

Pharmacokinetics and subjective effects of a novel oral LSD formulation in healthy subjects

This study characterised the pharmacokinetics of a novel oral LSD solution (100 μg) in 27 healthy subjects, finding first‑order elimination with a geometric mean Cmax ≈1.7 ng/mL at 1.7 h, t1/2 ≈3.6 h, measurable O‑H‑LSD but not nor‑LSD, and no sex differences. The subjective effects lasted about 8.5 h with peak effects at ≈2.5 h and were closely associated with plasma concentrations (EC50 ≈1.0 ng/mL for “good” and ≈1.9 ng/mL for “bad” effects).
