Acute psychotropic, autonomic, and endocrine effects of 5,6-methylenedioxy-2-aminoindane (MDAI) compared with 3,4-methylenedioxymethamphetamine (MDMA) in human volunteers: A self-administration study

In a non‑blinded study of six healthy volunteers, 5,6‑methylenedioxy‑2‑aminoindane (MDAI) produced subjective effects comparable with 125 mg MDMA and raised blood pressure and endocrine markers (cortisol, prolactin) but, unlike MDMA, did not increase heart rate or body temperature. MDAI was well tolerated and remained detectable in serum and urine for several days, warranting further clinical investigation of potential medicinal properties.

MDA, MDMA and other mescaline-like substances in the US military’s search for a truth drug (1940s to 1960s)

This review (2017) summarizes the history of the US military's attempt to operationalize psychotropic drugs, first scopolamine, and mescaline then later on MDMA and other derivatives, as a truth serum. These attempts were largely futile because scopolamine induced a delirium-like state of mind which raised doubt over the validity of the extracted information, whereas psychedelic substances produced excessive hallucinations, thought disturbance, and confusion, which hindered the interrogation process.

Health status of ayahuasca users

A review of 15 studies found that, beyond acute effects, ayahuasca use appears generally safe and may confer mental and physical benefits under certain conditions, but methodological biases and limited data preclude definitive conclusions. The authors call for further rigorous research to clarify ayahuasca’s long‑term health effects.

Metabolism and disposition of N,N-dimethyltryptamine and harmala alkaloids after oral administration of ayahuasca

Using HPLC‑ESI‑SRM‑MS/MS on 24‑hour urine from 10 healthy men given oral freeze‑dried ayahuasca (1.0 mg DMT/kg), the authors found <1% of DMT was excreted unchanged while ~50% appeared as indole‑3‑acetic acid, ~10% as DMT‑N‑oxide and 68% as total DMT plus metabolites, indicating MAO‑independent metabolic routes for DMT. Harmala alkaloids were mainly recovered as O‑demethylated conjugates with highly variable recoveries (9–65%), showing O‑demethylation/conjugation is important but not the only metabolic pathway.

A critical review of reports of endogenous psychedelic N, N-dimethyltryptamines in humans: 1955-2010

This paper critically reviews 69 studies (1955–2010) reporting endogenous N,N‑dimethyltryptamine, 5‑hydroxy‑DMT and 5‑methoxy‑DMT in human blood, urine and cerebrospinal fluid, evaluating the analytical methods and identification criteria used. The authors conclude that many early reports suffer from methodological shortcomings and inconsistent quantitation, and they offer recommendations to strengthen future research on endogenous psychedelic tryptamines.
